Sånger från 63° N is the name of the third album by the Swedish folk band Triakel. It was released on 23 June 2004 on Amigo Records; the songs are traditional Swedish folk songs from the province of Jämtland, though some are recent compositions. The text of I Jamtlann is in a Jämtland dialect; the rest of the songs are in Swedish. AllMusic noted the "chemistry between the members" and wrote that singer Emma Härdelin was best when presenting tragic songs; the reviewer for Helsingborgs Dagblad wrote that Triakel benefitted from the cooperation between the instrumental musicians and Härdelin's vocal performance. West Spring Gun

The West Spring Gun was a bomb-throwing catapult used by British and Australian forces during World War I. It was designed to throw a hand grenade in a high trajectory into enemy trenches, it consisted of a metal frame supporting a throwing arm powered by 24 metal springs. It was invented by Captain Allen West in 1915 and manufactured by the Reason Manufacturing Company of Brighton, granted a patent for the device on 19 October of that year. Although called a catapult, it was a hybrid of a trebuchet, it required a crew of five - three to compress the springs, one to load the bomb, one to fire as soon as the fuse was lit or the grenade pin was pulled. In tests, it could throw Mills bomb about 240 yd or a 7 lb projectile about 80 yd with a flight time of 6 or 7 seconds. In the field it threw a Jam Tin Grenade, No. 15 Ball grenade, No. 21 "Spherical" grenade or No. 28 chemical grenade, equipped with a longer fuse to ensure to reach the enemy trench before exploding. It was used in combat by, amongst others, the 50th Division and the 1st Canadian Division in the Second Battle of Ypres and by Australian forces in the Gallipoli Campaign.

It was considered to be large and cumbersome and "generally more unwieldy" than the Leach Trench Catapult. Many operators, including Captain West himself, lost fingers in the mechanism; the throw could be unpredictable, with the bomb sometimes landing near the thrower. Production of this and other trench catapults was halted in 1916, being replaced by the 2 inch Medium Trench Mortar and Stokes mortar. Poly (ADP-ribose) polymerase

Poly polymerase is a family of proteins involved in a number of cellular processes such as DNA repair, genomic stability, programmed cell death. The PARP family comprises 17 members, they have all different structures and functions in the cell. PARP1, PARP2, VPARP, Tankyrase-1 and -2 have a confirmed PARP activity. Others include PARP3, PARP6, TIPARP, PARP8, PARP9, PARP10, PARP11, PARP12, PARP14, PARP15, PARP16. PARP is composed of four domains of interest: a DNA-binding domain, a caspase-cleaved domain, an auto-modification domain, a catalytic domain; the DNA-binding domain is composed of two zinc finger motifs. In the presence of damaged DNA, the DNA-binding domain will bind the DNA and induce a conformational shift, it has been shown. This is integral in a programmed cell death model based on caspase cleavage inhibition of PARP; the auto-modification domain is responsible for releasing the protein from the DNA after catalysis. It plays an integral role in cleavage-induced inactivation; the main role of PARP is to detect and initiate an immediate cellular response to metabolic, chemical, or radiation-induced single-strand DNA breaks by signaling the enzymatic machinery involved in the SSB repair.

Once PARP detects a SSB, it binds to the DNA, undergoes a structural change, begins the synthesis of a polymeric adenosine diphosphate ribose chain, which acts as a signal for the other DNA-repairing enzymes. Target enzymes include DNA ligase III, DNA polymerase beta, scaffolding proteins such as X-ray cross-complementing gene 1. After repairing, the PAR chains are degraded via Poly glycohydrolase. NAD+ is required as substrate for generating ADP-ribose monomers, it has been thought that overactivation of PARP may deplete the stores of cellular NAD+ and induce a progressive ATP depletion and necrotic cell death, since glucose oxidation is inhibited. But more it was suggested that inhibition of hexokinase activity leads to defects in glycolysis. Basal PARP activity regulates basal bioenergetics. Note below that PARP is inactivated by caspase-3 cleavage during programmed cell death. PARP enzymes are essential in a number of cellular functions, including expression of inflammatory genes: PARP1 is required for the induction of ICAM-1 gene expression by cardiac myocytes and smooth muscle cells, in response to TNF.

The catalytic domain is responsible for Poly polymerization. This domain has a conserved motif, common to all members of the PARP family. PAR polymer can reach lengths of up to 200 nucleotides before inducing apoptotic processes; the formation of PAR polymer is similar to the formation of DNA polymer from nucleoside triphosphates. Normal DNA synthesis requires that a pyrophosphate act as the leaving group, leaving a single phosphate group linking deoxyribose sugars. PAR is synthesized using nicotinamide as the leaving group; this leaves a pyrophosphate as the linking group between ribose sugars rather than single phosphate groups. This creates some special bulk to a PAR bridge. One important function of PARP is assisting in the repair of single-strand DNA nicks, it binds sites with single-strand breaks through its N-terminal zinc fingers and will recruit XRCC1, DNA ligase III, DNA polymerase beta, a kinase to the nick. This is called base excision repair. PARP-2 has been shown to oligomerize with PARP-1 and, therefore, is implicated in BER.

The oligomerization has been shown to stimulate PARP catalytic activity. PARP-1 is known for its role in transcription through remodeling of chromatin by PARylating histones and relaxing chromatin structure, thus allowing transcription complex to access genes. PARP-1 and PARP-2 are activated by DNA single-strand breaks, both PARP-1 and PARP-2 knockout mice have severe deficiencies in DNA repair, increased sensitivity to alkylating agents or ionizing radiation. PARP activity measured in the permeabilized mononuclear leukocyte blood cells of thirteen mammalian species correlates with maximum lifespan of the species; the difference in activity between the longest-lived and shortest-lived species tested was 5-fold. Although the enzyme kinetics of the two enzymes were not different, human PARP-1 was found to have a two-fold higher specific automodification capacity than the rat enzyme, which the authors posited could account, in part, for the higher PARP activity in humans than rats. Lymphoblastoid cell lines established from blood samples of humans who were centenarians have higher PARP activity than cell lines from younger individuals, again indicating a linkage between longevity and repair capability.

These findings suggest. Thus, these findings support the DNA damage theory of aging, which assumes that un-repaired DNA damage is the underlying cause of aging, that DNA repair capability contributes to longevity; the tankyrases are PARPs that comprise ankyrin repeats, an oligomerization domain, a PARP catalytic domain. Anna Millikin

Anna Millikin was a teacher and author of the late 18th and early 19th centuries. She was one of the earliest Irish women to write Gothic novels and established the literary periodical the Casket or Hesperian Magazine. Millikin was born at Castlemartyr, County Cork from an Quaker family, she was the daughter of Elizabeth Battaley. Her grandfather, Robert Millikin, was a Belfast linen merchant of Scottish descent, he moved to the Boyle estate at Castlemartyr. At that point, the family became members of the established church. Millikin was one of the earliest women writers of Gothic fiction, she wrote at least four books. She founded and contributed to the literary periodical the Casket or Hesperian Magazine with her brother Richard Alfred Milliken, a poet, her subscribers of her novels included Lord Boyle, Countess Shannon, Lady Harriet Bernard of Bandon, the Freke family. The Critical Review of January 1803 reviewed her Plantagenet novel as follows: " In these two volumes of Mrs. Millikin, are contained the marriage of William, son to Robert duke of Normandy, with a daughter of the duke of Anjou.

Michael Madsen (footballer)

For other people with the same name, see Michael Madsen Michael Madsen is a Danish retired footballer who most prominently played professionally for Italian club AS Bari and VfL Wolfsburg in Germany. He is now the head coach of Brønshøj Boldklub. Madsen started his career at BK Avarta in the secondary Danish 1st Division league, where he won the 1994 Danish 1st Division Revelation of the Year award, he was introduced to the top-flight Danish Superliga championship, when he was brought to newly promoted club Akademisk Boldklub in June 1996. Under manager Christian Andersen, AB finished fifth in the 1997–98 Superliga season, with Madsen on the left flank, combining well with teammate Peter Knudsen in central midfield. In June 1998, Madsen and Peter Knudsen were both brought to Italian club AS Bari in a combined transfer deal worth DKK 15 million; when they were forced to compete for the same spot in the Bari starting line-up, Peter Knudsen returned to AB, while Madsen stayed three years at Bari.

He started out well for the club, but was benched by manager Eugenio Fascetti. Bari was relegated after the 2001 season, as his Bari contract ran out in June 2001, Madsen left the club, he moved to German club VfL Wolfsburg on a free transfer. In the opening weeks of the German Bundesliga championship season, Madsen suffered a back injury, struggled the remaining season to re-enter the starting line-up under manager Wolfgang Wolf without success. Following three league games for Wolfsburg, Madsen moved back to Denmark to play for Superliga club Farum BK under his former AB manager Christian Andersen in September 2002. Following one season at Farum, Madsen did not get his contract prolonged, decided to take a job in the sales department of SAV Danmark in July 2003. After a month away from football, he started playing for amateur club HIK in the Danish 2nd Division in August 2003, he helped the club win promotion for the 1st Division, which saw the club turn professional, before he moved to league rivals Lyngby BK in November 2005.

Due to an injured knee, he was forced to end his career in February 2007. Shortly after the news of his career stop, it was announced that he was to be associated with the newly founded Birkerød Sports College as a football coach, beginning in the summer of 2007. However, he chose to be new head coach at HIK, he only kept this job for a few months, because in June 2007 he was announced new sporting director at BK Fremad Amager. When his new club fired Jakob Friis-Hansen in May 2008 he became caretaker manager. After the season, he became manager of FC Amager, he had the job at Amager until March 2009. In June 2009 he was named the new manager of Vanløse IF. On 6 October 2011, he announced that he would not renew his contract with Vanløse, when it ended on 31 December 2011. In January 2012 he became assistant manager of Brønshøj Boldklub, he succeeded Erik Rasmussen as manager of Greve Fodbold on 1 January 2013.
